It really depends on what object you're attempting to Dedupe. I'd say generally though, my biggest saver has been using the Field Rules option. Definitely take a look at those options. For instance, if you're merging Accounts, make sure that your Trade/Industry field is set to 'combine values'. That goes for any multi-select picklist fields that you wouldn't want to lose the values for.
